Daru Spiritualist makes any targeted Cleric indestructible for the turn, which is mostly irrelevant — until you pair it with Ayli, Eternal Pilgrim or a free-sacrifice outlet and suddenly you have arbitrarily large toughness on demand. Outside that specific interaction, it's a 1/1 for two mana that does nothing on its own.

Daru Spiritualist sees essentially zero play outside Commander, where its combo potential with sacrifice loops gives it a narrow but real home. In Legacy and Vintage it's legal but competes with far more efficient two-drops and no one is building around it. Pauper is the only other format worth a mention — the combo pieces are all commons, but the loop lacks the redundancy and tutors that make it threatening. Commander is where Daru Spiritualist belongs: enough tutors exist to find it consistently, and Cleric-tribal shells give it a plausible home beyond the pure combo role.
