In Commander, Agatha's Champion is a build-around in aggressive red or Mardu strategies that deal early damage — the conditional size boost is easy to trigger when opponents start at 40 life and are already bleeding from early attacks. In 60-card competitive formats like Modern and Pioneer, five mana for a conditional 6/6 with trample is too slow without a dedicated shell to enable it reliably, and those formats have cheaper threats that don't ask as much of you. Legacy and Vintage have even less patience for it. Standard is where Agatha's Champion has the most plausible run if an aggressive red or Rakdos deck needs a top-end threat with reach, though it's competing against efficient five-drops that don't require setup.
